The following laboratory procedures for cultivating Trichoderma viride (Trichderma reesei) and assaying cellulases from ascomycetous fungi were developed by Dr. Elwin Reese and Dr. Mary Mandels at the U.S. Army Natick Laboratory from about 1955 until 1980. They were summarized in a laboratory handbook by Dr. Mandels in 1974. Most of these techniques have been published in several variations and some (such as the filter paper assay) have become standards for the field. Others now are outdated. The original copy of the Natick procedures manual that I received had been copied so many times that it had become illegible in parts, so I had it retyped in 1987 and am posting it here as much for historical reference as for contemporary use.
